Brain and Intelligence Seminar NO.42

On July 22, the Brain and Intelligence Seminar NO.42 was successfully held. This session featured Dr. Takashi Yamaguchi, Postdoctoral Fellow at the Institute for Translational Neuroscience, New York University, who presented a talk entitled “Maternal Brain Mechanisms Underlying Protective Aggression.”

In this seminar, Dr. Yamaguchi presented his latest work published in Nature in 2026. Lactating female mice exhibit intense aggression to protect their pups—a behavior known as maternal aggression—yet its rapid emergence after birth and swift decline when pups are absent have remained poorly understood. Dr. Yamaguchi‘s team revealed that the pathway from posterior amygdala cells expressing estrogen receptor alpha (PA^Esr1^) to ventrolateral VMH cells expressing neuropeptide Y receptor 2 (VMHvl^Npy2r^) plays a critical role in the rise and fall of maternal aggression. During lactation, PA-to-VMHvl^Npy2r^ synapses strengthen and VMHvl^Npy2r^ cell excitability increases, enabling heightened aggression. PA^Esr1^ neurons express abundant oxytocin receptors, allowing oxytocin to boost PA output; after pup removal, declining oxytocin levels reduce PA drive and dampen maternal aggression.

Based on these findings, Dr. Yamaguchi proposed a two-step hormonal control model: pregnancy-associated sex hormone surges remodel the aggression circuit, while lactation-associated neuropeptide signals (particularly oxytocin) gate circuit output in response to the need to protect offspring. This study reveals how multiple forms of plasticity in a defined neural circuit collectively implement the adaptive, need-based control of maternal aggression, providing a critical mechanistic framework for understanding reproductive state-dependent behavioral switching.
